Yes you can apply for PR from within Canada. You can apply for either
New Delhi or Buffalo. If applying to Buffalo you will need to have
valid US visa to enter in case of interview.

It means that work permit NOT EQUAL TO immigration visa.

You can apply for PR as long as you have atleast 12 months of VALID VISA

No problems joining her. You will not have healthcare coverage for 3
months though.
