There are both day and night busses and trains. The VIP night busses
tend to be very cold but they do provide blankets. The night train beds
offer a comfortable sleep unless you are very tall. Personally, I prefer
the overnight passage as the scenery from Bangkok until about 60 km from
Chiang Mai is almost identical; rice fields and banana plantations in
endless succession. But once you reach the mountains the views are
pretty and interesting to photograph as you pass through.

If you were to stop in Ayutthya for a day, stop in Sukkothai for a day,
then continue north I'd agree. But to make a non-stop trip, better to
take a night train or buss, or a quick 30 minute flight.

There are so many varied and interesting things in both cities, but I'd
give Bangkok three full days of sightseeing and Chiang Mai at least 4
full days. There is a much greater variety of sight to see and things to
do in the north.

Chiang Mai's airport is International. You can check your baggage, clear
immigration, and board in Chiang Mai, perhaps needing to change planes
in Bangkok, and collect your bags in S'pore with no problems. There are
several flights a day during December.
If you have more specific question, don't hesitate to e-mail me.
